Dissection of Nodule Development by Supplementation of \u3cem\u3eRhizobium leguminosarum\u3c/em\u3e biovar \u3cem\u3ephaseoli\u3c/em\u3e Purine Auxotrophs with 4-Aminoimidazole-5-Carboxamide Riboside
Purine auxotrophs of Rhizobium leguminosarum biovar phaseoli CFN42 elicit uninfected pseudonodules on bean (Phaseolus vulgaris L.). Addition of 4-aminoimidazole-5-carboxamide (AICA) riboside to the root medium during incubation of the plant with these mutants leads to enhanced nodule development, although nitrogenase activity is not detected. Nodules elicited in this manner had infection threads and anatomical features characteristic of normal nodules, such as peripheral vasculature rather than the central vasculature of the pseudonodules that were elicited without AICA riboside supplementation. Although 105 to 106 bacteria could be recovered from these nodules after full development, bacteria were not observed in the interior nodule cells. Instead, large cells with extensive internal membranes were present. Approximately 5% of the normal amount of leghemoglobin and 10% of the normal amount of uricase were detected in these nodules. To promote the development of true nodules rather than pseudonodules, AICA riboside was required no later than the second day through no more than the sixth day following inoculation. After this period, removal of AICA riboside from the root medium did not prevent the formation of true nodules. This observation suggests that there is a critical stage of infection, reached before nodule emergence, at which development becomes committed to forming a true nodule rather than a pseudonodule
Infection of Soybean and Pea Nodules by \u3cem\u3eRhizobium\u3c/em\u3e spp. Purine Auxotrophs in the Presence of 5-aminoimidazole-4-Carboxamide Riboside
Purine auxotrophs of various Rhizobium species are symbiotically defective, usually unable to initiate or complete the infection process. Earlier studies demonstrated that, in the Rhizobium etli-bean symbiosis, infection by purine auxotrophs is partially restored by supplementation of the plant medium with 5-amino-imidazole-4-carboxamide (AICA) riboside, the unphosphorylated form of the purine biosynthetic intermediate AICAR. The addition of purine to the root environment does not have this effect. In this study, purine auxotrophs of Rhizobium fredii HH303 and Rhizobium leguminosarum 128C56 (bv. viciae) were examined. Nutritional and genetic characterization indicated that each mutant was blocked in purine biosynthesis prior to the production of AICAR. R. fredii HH303 and R. leguminosarum 128C56 appeared to be deficient in AICA riboside transport and/or conversion into AICAR, and the auxotrophs derived from them grew very poorly with AICA riboside as a purine source. All of the auxotrophs elicited poorly developed, uninfected nodules on their appropriate hosts. On peas, addition of AICA riboside or purine to the root environment led to enhanced nodulation; however, infection threads were observed only in the presence of AICA riboside. On soybeans, only AICA riboside was effective in enhancing nodulation and promoting infection. Although AICA riboside supplementation of the auxotrophs led to infection thread development on both hosts, the numbers of bacteria recovered from the nodules were still 2 or more orders of magnitude lower than in fully developed nodules populated by wild-type bacteria. The ability to AICA riboside to promote infection by purine auxotrophs, despite serving as a very poor purine source for these strains, supports the hypothesis that AICAR plays a role in infection other than merely promoting bacterial growth

After an introduction to diabetes and glucose sensing, this review analyses state of the art and pipeline devices; in particular their user friendliness and technological advancement. Extended Photometry for the DEEP2 Galaxy Redshift Survey: A Testbed for Photometric Redshift Experiments
This paper describes a new catalog that supplements the existing DEEP2 Galaxy
Redshift Survey photometric and spectroscopic catalogs with ugriz photometry
from two other surveys; the Canada-France-Hawaii Legacy Survey (CFHTLS) and the
Sloan Digital Sky Survey (SDSS). Each catalog is cross-matched by position on
the sky in order to assign ugriz photometry to objects in the DEEP2 catalogs.
We have recalibrated the CFHTLS photometry where it overlaps DEEP2 in order to
provide a more uniform dataset. We have also used this improved photometry to
predict DEEP2 BRI photometry in regions where only poorer measurements were
available previously. In addition, we have included improved astrometry tied to
SDSS rather than USNO-A2.0 for all DEEP2 objects. In total this catalog
contains ~27,000 objects with full ugriz photometry as well as robust
spectroscopic redshift measurements, 64% of which have r > 23. By combining the
secure and accurate redshifts of the DEEP2 Galaxy Redshift Survey with ugriz
photometry, we have created a catalog that can be used as an excellent testbed
for future photo-z studies, including tests of algorithms for surveys such as

Electrochemical peroxidation of contaminated water and assessment of the toxicity using existing and novel bioassays
The treatment of wastewater and monitoring of its toxicity is essential before discharging it to the environment. This study focuses on assessing the toxicity of wastewater following treatment using the electrochemical peroxidation process (ECP). Three categories of low-cost toxicity assays were used based on plant cells, microorganisms and invertebrates, all of which do not require ethical approval or a special licence. In addition, a novel cost-effective device was developed for assessing wastewater toxicity at low concentrations. In Chapter 2, the problem of high turbidity of the ECP-treated samples is addressed, by accelerating particulate settling of the existent compounds. This was achieved by storing the ECP treated samples at low temperatures. X-ray Surface Brightness Profiles of Active Galactic Nuclei in the Extended Groth Strip: Implications for AGN Feedback
Using data from the All Wavelength Extended Groth Strip International Survey
(AEGIS) we statistically detect the extended X-ray emission in the interstellar
medium (ISM)/intra-cluster medium (ICM) in both active and normal galaxies at
0.3 <= z <= 1.3. For both active galactic nuclei (AGN) host galaxy and normal
galaxy samples that are matched in restframe color, luminosity, and redshift
distribution, we tentatively detect excess X-ray emission at scales of 1--10
arcsec at a few sigma significance in the surface brightness profiles. The
exact significance of this detection is sensitive to the true characterization
of Chandra's point spread function. The observed excess in the surface
brightness profiles is suggestive of lower extended emission in AGN hosts
compared to normal galaxies. This is qualitatively similar to theoretical
predictions of the X-ray surface brightness profile from AGN feedback models,
where feedback from AGN is likely to evacuate the gas from the center of the
galaxy/cluster. We propose that AGN that are intrinsically under-luminous in
X-rays, but have equivalent bolometric luminosities to our sources will be the
ideal sample to study more robustly the effect of AGN feedback on diffuse
ISM/ICM gas.Comment: Accepted in PAS
Measurement of an AGN Central Mass on Centiparsec Scales: Results of Long-Term Optical Monitoring of Arp 102B
The optical spectrum of the broad-line radio galaxy Arp 102B has been
monitored for more than thirteen years to investigate the nature of the source
of its broad, double-peaked hydrogen Balmer emission lines. The shape of the
lines varied subtly; there was an interval during which the variation in the
ratio of the fluxes of the two peaks appeared to be sinusoidal, with a period
of 2.16 years and an amplitude of about 16% of the average value. The variable
part of the broad H-alpha line is well fit by a model in which a region of
excess emission (a quiescent ``hot spot'') within an accretion disk (fitted to
the non-varying portion of the double-peaked line) completes at least two
circular orbits and eventually fades. Fits to spectra from epochs when the hot
spot is not present allow determination of the disk inclination, while fits for
epochs when it is present provide a measurement of the radius of the hot spot's
orbit. From these data and the period of variation, we find that the mass
within the hot spot's orbit is 2.2 +0.2/-0.7 times 10^8 solar masses, within
the range of previous estimates of masses of active galactic nuclei. Because
this mass is determined at a relatively small distance (~1000 AU) from the
central body, it is extremely difficult to explain without assuming that a
supermassive black hole lies within Arp 102B. The lack of any systematic change
in the velocity of the blue peak over time yields a lower limit on the combined
mass of the two bodies in a binary black hole model like that of Gaskell (1983)

A revised Cepheid distance to NGC 4258 and a test of the distance scale
In a previous paper (Maoz et al. 1999), we reported a Hubble Space Telescope
(HST) Cepheid distance to the galaxy NGC 4258 obtained using the calibrations
and methods then standard for the Key Project on the Extragalactic Distance
Scale. Here, we reevaluate the Cepheid distance using the revised Key Project
procedures described in Freedman et al. (2001). These revisions alter the zero
points and slopes of the Cepheid Period-Luminosity (P-L) relations derived at
the Large Magellanic Cloud (LMC), the calibration of the HST WFPC2 camera, and
the treatment of metallicity differences. We also provide herein full
information on the Cepheids described in Maoz et al. 1999. Using the refined
Key Project techniques and calibrations, we determine the distance modulus of
NGC 4258 to be 29.47 +/- 0.09 mag (unique to this determination) +/- 0.15 mag
(systematic uncertainties in Key Project distances), corresponding to a metric
distance of 7.8 +/- 0.3 +/- 0.5 Mpc and 1.2 sigma from the maser distance of
7.2 +/- 0.5 Mpc. We also test the alternative Cepheid P-L relations of Feast
(1999), which yield more discrepant results. Additionally, we place weak limits
upon the distance to the LMC and upon the effect of metallicity in Cepheid

Anomalous scaling and Lee-Yang zeroes in Self-Organized Criticality
We show that the generating functions of avalanche observables in SOC models
exhibits a Lee-Yang phenomenon. This establishes a new link between the
classical theory of critical phenomena and SOC. A scaling theory of the
